| 2023 | Tractable and Near-Optimal Adversarial Algorithms for Robust Estimation in Contaminated Gaussian ModelsabstractConsider the problem of simultaneous estimation of location and variance matrix under Huber's contaminated Gaussian model. First, we study minimum $f$-divergence estimation at the population level, corresponding to a generative adversarial method with a nonparametric discriminator and establish conditions on $f$-divergences which lead to robust estimation, similarly to robustness of minimum distance estimation. More importantly, we develop tractable adversarial algorithms with simple spline discriminators, which can be defined by nested optimization such that the discriminator parameters are determined by maximizing a concave objective function given the current generator. The proposed methods are shown to achieve minimax optimal rates or near-optimal rates depending on the $f$-divergence and the penalty used. This is the first time such near-optimal error rates are established for adversarial algorithms with linear discriminators under Huber's contamination model. We present simulation studies to demonstrate advantages of the proposed methods over classic robust estimators, pairwise methods, and a generative adversarial method with neural network discriminators. | 2022 | On Loss Functions and Regret Bounds for Multi-Category ClassificationabstractWe develop new approaches in multi-class settings for constructing loss functions and establishing corresponding regret bounds with respect to the zero-one or cost-weighted classification loss. We provide new general representations of losses by deriving inverse mappings from a concave generalized entropy to a loss through a convex dissimilarity function related to the multi-distribution$f$-divergence. This approach is then applied to study both hinge-like losses and proper scoring rules. In the first case, we derive new hinge-like convex losses, which are tighter extensions outside the probability simplex than related hinge-like losses and geometrically simpler with fewer non-differentiable edges. We also establish a classification regret bound in general for all losses with the same generalized entropy as the zero-one loss, thereby substantially extending and improving existing results. In the second case, we identify new sets of multi-class proper scoring rules through different types of dissimilarity functions and reveal interesting relationships between various composite losses currently in use. We also establish new classification regret bounds in general for multi-class proper scoring rules and, as applications, provide simple meaningful regret bounds for two specific sets of proper scoring rules. These results generalize, for the first time, previous two-class regret bounds to multi-class settings. Zhiqiang Tan, Xinwei Zhang 0004 |

| 2018 | Learning Trans-Dimensional Random Fields with Applications to Language ModelingabstractTo describe trans-dimensional observations in sample spaces of different dimensions, we propose a probabilistic model, called the trans-dimensional random field (TRF) by explicitly mixing a collection of random fields. In the framework of stochastic approximation (SA), we develop an effective training algorithm, called augmented SA, which jointly estimates the model parameters and normalizing constants while using trans-dimensional mixture sampling to generate observations of different dimensions. Furthermore, we introduce several statistical and computational techniques to improve the convergence of the training algorithm and reduce computational cost, which together enable us to successfully train TRF models on large datasets. The new model and training algorithm are thoroughly evaluated in a number of experiments. The word morphology experiment provides a benchmark test to study the convergence of the training algorithm and to compare with other algorithms, because log-likelihoods and gradients can be exactly calculated in this experiment. For language modeling, our experiments demonstrate the superiority of the TRF approach in being computationally more efficient in computing data probabilities by avoiding local normalization and being able to flexibly integrate a richer set of features, when compared with n-gram models and neural network models. | 2015 | Deep Sequencing of Protease Inhibitor Resistant HIV Patient Isolates Reveals Patterns of Correlated Mutations in Gag and ProteaseabstractWhile the role of drug resistance mutations in HIV protease has been studied comprehensively, mutations in its substrate, Gag, have not been extensively cataloged. Using deep sequencing, we analyzed a unique collection of longitudinal viral samples from 93 patients who have been treated with therapies containing protease inhibitors (PIs). Due to the high sequence coverage within each sample, the frequencies of mutations at individual positions were calculated with high precision. We used this information to characterize the variability in the Gag polyprotein and its effects on PI-therapy outcomes. To examine covariation of mutations between two different sites using deep sequencing data, we developed an approach to estimate the tight bounds on the two-site bivariate probabilities in each viral sample, and the mutual information between pairs of positions based on all the bounds. Utilizing the new methodology we found that mutations in the matrix and p6 proteins contribute to continued therapy failure and have a major role in the network of strongly correlated mutations in the Gag polyprotein, as well as between Gag and protease. Although covariation is not direct evidence of structural propensities, we found the strongest correlations between residues on capsid and matrix of the same Gag protein were often due to structural proximity. This suggests that some of the strongest inter-protein Gag correlations are the result of structural proximity. Moreover, the strong covariation between residues in matrix and capsid at the N-terminus with p1 and p6 at the C-terminus is consistent with residue-residue contacts between these proteins at some point in the viral life cycle.
